“The present norm has as main objective, the determination of Staphylococcus coagulase and non
producers, for sanitary control of water pool analysis.
Staphylococcus is saprofitic bacteria, from normal flora of Human and therefore can de consider as good
indicators for human contamination, having potential pathogenic strains.”
Filtrar 100 mL da amostra de Água, ou as suas diluições, com filtro em
MCE de 0,45 µm e 47 mm ∅ (ref. 1080200), e transferir o filtro para
placa de meio Mannitol Salt Agar (ref. 610029; 10030)
Incubar a 37±1ºC/48h
Contar colónias características, colónias amarelas ou brancas,
envolvidas ou não por um halo amarelo e seleciona-se 3 colónias de
cada tipo para isolar em meio não seletivo, Nutrient Agar (ref. 610036;
10044; 412190) e incubar a 37±1ºC/24h

Regenerar meio semi-sólido de carne e levedura (M.E.V.A.G),
previamente distribuído em tubos, durante 15 min, manter tubos
a 45±1ºC. Inocular uma colónia em profundidade. Incubar a
37±1ºC/24h e verificar o crescimento em todo o tubo
(Estafilococos são bactérias aeróbias e anaeróbias facultativas)
